Briefly, pre-stabilized whole blood samples were thawed at room temperature and mixed vigorously in a 15 mL conical tube, chloroform was added to the sample, and centrifuged. The aqueous phase was transferred to another tube and the RNA was precipitated with ethanol.
The mixture was passed through a QIA-shredder spin column and samples were processed by another step of column purification and in-column RNase-free DNA digestion according to the supplier’s recommendations. Purification of purified RNA was performed in RNase-free water treated with diethyl pyrocarbonate (Sigma, St. Louis). RNA integrity was assessed on a Biolyzer 2100 instrument (Agilent Technologies, USA), using samples with RNA integrity number > 6. RNA was converted to cDNA using Superscript II reverse transcriptase kit (Invitrogen, USA) , briefly, 2.5 μg of total RNA according to manufacturer’s protocol in Biometra Gradient model T thermal cycler (Thermo Corp., Germany) was written in reverse. cDNA was treated with DNA-free RNase H, and cDNA aliquots were prepared at a concentration of 25 ng/μl for quantitative PCR. inwiefern till knoke
